    True Sunless Tanning


    The discovery of a molecular switch that turns off the natural process of skin pigmentation may lead to a novel way of protecting the skin -- activating the tanning process without exposure to cancer-causing UV radiation. In their report in the journal Genes & Development, researchers from the Massachusetts General Hospital (MGH) Cutaneous Biology Research Center (CBRC) describe how blocking the action of this switch -- an enzyme called PDE-4D3 -- in the skin of mice led to a significant increase in melanin production.

    "The primary goal of inducing melanin production in human skin would be prevention of skin cancer, since all the common forms are known to be associated with UV exposure, " explains David Fisher, MD, PhD, director of the hospital's Department of Dermatology and an investigator at the MGH CBRC, who led the study. "Not only would increased melanin directly block UV radiation, but an alternative way to activate the tanning response could help dissuade people from sun tanning or indoor tanning, both of which are known to raise skin cancer risk."

    In 2006 Fisher's group showed that the metabolic pathway leading to UV-induced pigmentation is controlled by cyclic AMP (cAMP), a molecule known to regulate many important cellular processes by carrying messages from the cell surface to internal target molecules. Using a strain of transgenic mice with red hair and melanocytes in their epidermis -- common mice have none of these melanin-producing cells in the outer skin layer -- they found that inducing cAMP production in the animals' skin led to significant pigmentation. But since the drug used in that study cannot penetrate human skin, they needed to investigate an alternative approach.

    Because most drugs act by blocking rather than stimulating their target molecules, better defining the pathway leading from UV exposure to melanin production could identify a step limiting melanin expression that, if suppressed, would increase production of the pigment. The strength and duration of the signals carried by cAMP are controlled by PDE enzymes, which break down the molecule after its message is delivered. Detailed analysis of the melanin expression pathway identified PDE-4D3 as the regulator of cAMP activity in melanocytes. The transcription factor activated by cAMP induces production of both melanin and PDE-4D3, and the enzyme in turn modulates the pigmentation process by breaking down cAMP.

    The researchers confirmed role of PDE-4D3 in controlling melanin expression by applying several agents that block PDE production to the skin of the transgenic mice with epidermal melanocytes. After five days of treatment, the animals' skin had darkened significantly, while treament of control mice with no epidemal melanocytes produced no effect.

    "Although PDE enzymes degrade cAMP within all cells, different members of this enzyme family are active in different types of cells," Fisher explains. "We showed that PDE-4D3 is particularly important within melanocytes, and while the enzyme may have a role in other cells, a blocking drug that is applied directly to the skin would probably have limited effects in other tissues." Additional research is needed to identify drugs that penetrate human skin and safely block PDE-4D3, he notes, and his team has already starting searching for such agents.

    Those who love to tan but are not fond of being exposed to sun will surely be happy about this! This information will be a great help to those who want to have a beautiful tan. Thank you for this very detailed information. I can see that you really have great articles regarding tanning/sun exposure and the likes.

    While I am really interested about this, I am not so sure whether I am ready to give melanotan up as I am very satisfied with the result. However, I might try to suggest this to some of my friends who might be interested in trying this method. I hope it will work great!

    I agree with you. Melanotan is too good to stop using. It delivers the kind of tan that I was just wishing for before. ANyway, thenk you for sharing this to us @Semin. For sure, many people will love this post as well.

    I also don't think that the OP will stop people from using MT either to be honest.

    Millions of people use MT and are more than happy with the results they get so as with anything, if you find one thing that works for you, then you make sure you stick with it.

    The only effective sunless tanning I know is with the use of a tanning bed. Though you don't have to expose yourself under the heat of the sun, you'll still be exposed to UV. Well, it might seem like the same thing and the only difference is that tanning bed will most likely give you even tan.

    That makes sense. Lol. You still get a nice tan even when you don't go out. Even in winter (when there's no sun around), you can still be tan if you want to. Thus, it is really the bets sunless tanning for me as well.

    An increase in melanin production will definitely make your skin darker. I wonder how effective this so-called "sunless tanning" is. Well, since I am already satisfied with melanotan and tanning bed, I guess I won't be able to try it. I am actually scared to try new products only to be disappointed by the results. Since I am already satisfied, I don't want to look for anything else.

    After reading this, I become interested on what kind of drug would turn your skin dark without being exposed to UV. If I am the user, I'd definitely freak out how my skin changes after taking this drug. Lol.

    This is the first time I've heard about this sunless tanning. I didn't even think it is possible. Well, I guess I have to do my research. While the article posted in this thread is surely helpful, I still need more information regarding this. I have to know more before i decide to give it a try.

    When I want to do sunless tanning I usually use products like Jergens Natural Glow. this is a good product and you can get some real darkening effects from it. the only problem iwht this product is of course it doesn't last forever, and their is the streakiness factor. I am not that desperate to have a tan that I am willing to ingest something to give me a tan. However, I know a lot of people on here like it, so for those that do, I think it is a good thing and something that can be helpful to them.

    I had friends who really spent long hours outside the sun to get a tan for their skin. It just happened that I am not one of them who want to have a tan color for my skin. Maybe if they will know this information they will surely research and check about this thing. But I will make it sure that I will share this information with them and thanks for sharing!

    Actually, tanning here in my country is not that popular. People tend to whiten their skin. So even if many people here have beautiful, tanned skin, they opt to whiten it. But this is a great article and certainly a better option than exposing yourself in the harsh sun just to get that certain tan you want. I do know some people from some blogs and channels that use lotions to achieve the tan that they want. So this would be an easier option for them and for those wanting to be tan.

    But honestly, getting some sun is still the best way to go, just as long as you don't overdo it. You get vitamins and nutrients from the sun, which is really good for our skin.
