You'll likely find after your first 500mcg (0.5mg) dose that you'll have 4-6 hour onset, with anywhere between about 8-48 hours of effect.
The following week at the same dosage you'll have similar onset, but maybe 90% of the effect.
Keep at 500mcg until it's about 50% as long, then step up to 750mcg - this should be enough to bring you right back up to 100%.
Rinse and repeat until you find your dose that is consistent and you're happy with.
It took me about 6 months to reach 1500mcg (1.5mg) at which point it starts to plateau - injecting more doesn't help, but the effects are consistent with 4 hours onset and 48 hour effect.
Other people report longer and shorter times, so it may vary between people.
There's also others who jump in at the deep end at 1-1.5mg, only to find that a few weeks down the line when they need to up the dose it doesn't work - and they've ruined it for themselves. A similar deal happens for people who use it daily. A cooldown period of 6-12 months has apparently lowered some people's tolerance so they can start again, but there's no actual figures on this.
